As we know our environmental conditions are not capable to store the all type of seeds in this earth . Seeds may be get degenerated or may be get infertile. So because of this condition. The term name 'CRYOPRESERVATION' is used. Which mean
Long term preservation of cells or seeds is used by storing them into the liquid nitrogen at -196°C . This process is called cryopreservation.
It is the most successful method of preservation culture cells, and animal sperms and embroys. Firstly this process used for storing the seeds. After that for the new biotechnology research happened. It's is now using for the store of healthy male sperm of any animal and ovum of the female.
In some cases the zygote is also store by this process. To easy for the researcher to move one place to another. For the transportations.
Why we use cryopreservation?
It's is desirable due to saveral reasons , including the risk of genetic variation, contamination, phenotypic risk etc.
Process of CRYOPRESERVATION
1- select the authentic cell line to be preserved which is free from contamination.
2- cell line cloned continuously and a single cell line clone is selected after that it characterised, multiplied and preserved.
3- the cells are suspended in a suitable medium at a high cell density, and a cryoprotectant is added .
4- cryoprotectant is such as glycerol or DMSO (dimethyl sulfoxide) is added prior to freezing. It is the most widely used cryoprotectant at concentrations ranging from 5 to 15 percentage.
Material
Laboratories generally use 'polypropylene ampoules' of 1.2ml , but the cell bank and respositories prefer glass ampoules for seed stocks.
Glass ampoules is used only by the experienced workers and they perfectly and quickly sealed in a gas oxygen flame. The workers do this and make sure there is no leakages.
The level of liquid nitrogen and the temperature in the cryofreezer is regularly monitored, and it's is ensure that the temperature remains below upto -170°C.
When it's time to use these preserved object. The forzen ampoules thawed as quickly as possible by immersing them in warm in 37°C water in a water bath.
